By using an analogue of lactate [2-hydroxy-3-butenoic acid (vinylglycolate; VG)] which is transported by a lactate transport protein and then oxidized inside of the vesicles to a reactive species (2-keto-3-butenoic acid), it is shown that the rate limiting step for covalently labeling vesicles is transport.
